It happens with just about every new character who is introduced into a story where the main and primary supporting cast has been firmly established. Questions like: who are they really; what is their backstory; can they be trusted; and so on always abound. Of course, when the character introduced already has a slightly-suspicious background and manages to turn a few supposed-rules on their heads, well, it's safe to say that there will be even more questions and excitement raised!

Godaime Mizukage & Yonbi Jinchuuriki Related?

Kisame remarks on the yonbi jinchuuriki's ability to use youton techniques Despite the fact that the Yonbi jinchuuriki was a shinobi of Iwagakure and the Godaime Mizukage is clearly from Kirigakure there are those that suspect that the two are related given their similarity in abilities.

In chapter 353 of the manga, the Yonbi jinchuuriki is seen to have been defeated and captured by the Akatsuki. Itachi, concerned that the jinchuuriki may die on them, is of the opinion that Kisame may have been too rough on him. Kisame, however, disagrees, and mentions that Roshi, the Yonbi jinchuuriki, is unlike the other jinchuuriki because of his ability to use Youton techniques. Choujuuro, one of the Mizukage's guards, implicitly states that her Youton abilities are a bloodline. So are they related? While both Roshi and the Mizukage use Youton it should be noted that there is a distinct difference in their techniques.

Roshi's abilities are referred to as "Youton" with a fire-based kanji:

Mizukage's abilities are referred to as "Youton" with a water-based kanji:

Youton itself is spelled the same when written in romanji, however, when written in kanji the two techniques are spelled differently with very significant changes that imply a difference in how they are made up.

Choujiro calls the Mizukage's technique a bloodline named Youton Keep in mind that this is not the first time someone without a bloodline was able to wield bi-elemental techniques as well. If you recall, Haku's Hyouton techniques were a bloodline that came naturally to him and was a result of him combining his wind and water-natured chakra together to create ice. Salamandar Hanzo did not have a bloodline but he was reknown for his Hyouton techniques as well. Kishimoto has demonstrated that unlike bloodlines that physically affect or alter the body (ie: Shikotsumyaku, Byakugan, Souma no Kou) bi-elemental bloodlines can be re-created by non-bloodline holders. However, given that those known to have been able to successfully do so were monstrously powerful themselves, it is not likely that just anyone will be able to set out and accomplish this feat. This makes the bloodline-holders who can do so naturally with ease that much more devastating. More than likely it takes a great deal of elemental manipulation skills, ability, and chakra to do so effectively without the aid of a bloodline.

Current Conclusion:
The 5th Mizukage is is related to Roshi the Yonbi Jinchuuriki! - Not at all.
